Disruption of addiction memory reconsolidation by β-AR inhibition after cocaine self-administration induced memory retrieval

摘 要:Drug relapse is a major clinical problem mainly caused by maladaptive drug memory.Disruption of drug memory would reduce relapse susceptibility.In this study,we used drug retrieval procedure to investigate the role ofβ-adrenergic receptor(β-AR)in addiction memory reconsolidation.Rats were injected propranolol(β-AR antagonist),betaxolol(β1-AR antagonist),ICI-118,551 HCl(β2-AR antagonist)immediately after drug memory retrieval with self-administration of cocaine(3mg/kg).Our data showed thatβ-AR antagonist especiallyβ1-AR antagonist disrupted cocaine seeking behavior 24 h after retrieval and cocaine priming induced memory reinstatement after extinction procedure.On the other hand,β2-AR antagonist had no effect on drug seeking 24 h after retrieval.The data above suggest a potential role ofβ1-AR in addiction memory reconsolidation following cocaine self-administration induced memory retrieval.Blockade ofβ1-AR after drug memory retrieval may be a promising treatment for cocaine relapse.
