摘 要:Progesterone has been shown to be a neuroprotective agent for ischemic damage.Progesterone reduces infarct volume and promotes behavioral recovery in ischemic stroke models.However,the neuroprotective mechanisms of progesterone remain unclear.Microcirculation in peri-infarct area is one of the critical targets in stroke therapy.We hypothesize that progesterone may improve microcirculation in ischemic penumbra and postpone expansion of infarct border.Using transcranial two-photon microscopic imaging technique and transgenic YFP fluorescence mice,we monitored effect of progesterone on the expansion of infarct border,cerebral blood flow,dendritic spine stability in peri-infarct region after cerebral ischemia.Our results suggested that progesterone decreased the expansion of neuronal infarct border within three days,especially in the first 24 hours.The expansion of infarct border was reduced by 34.59%at the first 24 h,and was reduced by 47.22%at 72 h following ischemia in progesterone group compared with the control.Change of infarct border was not significant after three days.Our data suggested that dendrites and spines were damaged within 300μm from the ischemic border,and spine elimination was negatively correlated with the distance to the infarct border.Progesterone decreased the spine elimination rate in peri-infarct region 100μm away from the ischemic border,but spine formation rate was not increased.Two photon line-scanning revealed that progesterone significantly delayed the reduction of cerebral blood flow in peri-infarct region after ischemic brain injury.The red blood cell velocity and flux gradually decreased in peri-infarct region after ischemia,and progesterone postponed the reduction of red blood cell velocity and flux at 1 h after treatment.Evans blue assay indicated that progesterone maintained the integrity of blood brain barrier.In conclusions,our results suggested that progesterone limited the progression of ischemic border and increased the stability of dendritic spines.The neuroprotective effe
